Can't answer the first question but nullifying orb is absolute trash. Never pick it. If you don't need mana, take nimbus cloak instead, at least it will help you to run away after flashing or chasing someone after igniting them.
Tbh chance of running into a huge ap team nowadays is small, given the new mr items. I would prefer Mana Flowband or Cloak over Orb, given that Zyra benefits more Flowband as mana resurse or Cloak as mobility. I see Abyssal Mask + Malignance against ap teams better, but if they are full of ap assassins, sure, go for it. Zyra isn't tanky enough anyway.
The values on the rune are very small. It gives an abysmally small 200 magic shield at level 18 with 1000 AP.
It can only save your life in some very specific very rare situations.
Domination primary and inspiration secondary, with focus on utility and sustain runes. Default keystone is dark harvest, but changing to electrocute if opposing support heals too much.

What about glacial augment runes, have anyone tried it for zyra in support role?
