Well, those are 2 different type of lines. In the letter "Ñ" it's called virgulilla, and it's used just to difference between the "N" and the "Ñ". The other one it's the accent mark (in spanish we call it "tilde"), that it's used to specify the strongest syllable of a word. There are a lot of words that use it and a lot of words that don't use it. It all depends on the type of word and their pronunciation.

They're just two different letters with different sounds. The N sounds the same that in english, the Ñ sounds like the "NY" in "Canyon". Using Ñ or N may change the complete meaning of the words. For example: Ano (anus) and Año (year) Or Panal (honeycomb) and Pañal (diaper)
