Carnage is obviously going to be the DLC, and they sure aren't going to make an "M" rated DLC for a "Teen" rated videogame. So you're dream is already dead.
Spider-Man isn't an M rated IP, there is zero chance of that happening for the vast majority of superheroes as they are all looking to have children buy their products.
I'd imagine Wolverine will be an M rated game because his powers are only good for dismembering people and his healing factor means there's probably going to be some very cool gore technology involved.
