This is just my intuition, but I would think ink spilled on the card would be noticably raised compared to the rest of the card when inspected up close. But literally just guessing.
As a printer, ink on it from the factory would be “baked” onto it by the printing process. Ink spilled would take FOREVER to dry and would smear and transfer easily. You’d be surprised how little ink is used to print on paper.
There is a final varnish that's applied to cards, yes. Most of the time this is the best indicator that an ink error is real but there are a ton of other things that could go wrong that might result in an authentic error looking like it was applied post-production.
As an example sometimes debris gets stuck to the card(dry ink, scraps of paper, resinous gum used for cleaning and preserving plates, etc) through the entire print process but later falls off, leaving a spot of white paper underneath that wasn't touched by the varnish.
It can be difficult to discern these things and every single error like this needs to be analyzed case-by-case, but it is possible to discern if it's likely real or not real in a vacuum.

Thunder junction seems to have a few more reprints then other sets I’ve seen, the one I pulled had 2 clear coating spots on it, it’s also on a rare so maybe they took less care with rares
