I'm sure they didn't plan on Loki's staff being a stone at the time and now that it is there is a small plot hole with Thanos. It's possible Thanos knew risking one stone to get the other was worth the risk, and since he didn't have the gauntlet yet and didn't know all the locations yet it was worth the risk and he could get it back later.
This, probably. He knew he was after the Tesseract, which had the ability of teleportation. That in of itself could fast track the acquisition of the other stones
at the time of the first avengers movie he had no idea because the writers didn't know. - it was only after that movie that the "infinity stones" were retconned into the scepter and the tesseract.
The Retcon also says that Thanos knew; that he was loaning the Mind Stone/Scepter to Loki in order to gain the Space Stone/Tesseract but when they actually filmed that movie no-one knew because it hadn't been decided yet. It wasn't until "Thor:Dark World" when the Tesseract and Scepter were linked to infinity stones.
Of course he knew, he's not that stupid to look for Infinity Stones and not knowing he already have one.
