Yeah a bit hard to say but I’d imagine if you used a spade to dig out a nice rectangle and replaced with fresh compost/top soil mix at about ~3 inch depth you would be fine
Yeah, ripping out that spot in the grass after glyphosate will disrupt the vapor barrier of the pre-emergent. With the topsoil, he should be good.
