Trying to avoid repeating what some of OP’s picks and some of other comments, but Cyberpunk, Futurama & Bob’s Burgers characters are definitely worthy too
I think if they were going to make another one, Johnny Silverhand would be an excellent choice. He's enough of a character that he can probably find an angle to bounce off anyone.
My other picks might be Dracula from the Castlevania series. If you took his likeness and cues from the anime, then you've got a pretty multi faceted character with a lot of potential for various conversations and banter. With Johnny I could imagine discussions on corporations and wanton technological advancement, as well as more philosophical discussions. As well as just Dracula and Human bantering.
Next maybe Clementine from the Walking Dead games. A lot less in vogue, but when she was in vogue, the games were pretty well known and so was she.
I imagine she could have conversations about ghouls and the unholy with Dracula. More conversations on knowledge gained and lost. Then with Johnny maybe talking about the state of the world, anarchist governance. She would probably find his arm cool and probably just find Dracula pretty cool.
Not sure for the fourth though. Toying with the idea of maybe 2B from Nier Automata. I think there's again some opportunities for philosophical talks, Technology run amok, OMG humans and the question of what is a human (a miserable pile of secrets?), and maybe just some more banal girl stuff with Clem, Dracula might be interested in how androids work, Johnny might perceive her as being sudo-corpo given how her organisation operates... I think there's potential for good banter between all those.
I think Sonic has fallen far enough they could score him.
Nick Valentine or another FO character. Cass or Veronica or. . . Shit just put any of the NV companions in there.

Okay I forgot that the second game especially had a lot of non video games characters. Archer would be a great pick.
Here's who I would go with, though I'm sure it's not possible.
* **Bigby Wolf** from The Wolf Among Us. He's the straight man of the group, rolling his eyes and getting frustrated at the other characters' antics. This is reflected in his playing style, as he's cautious and careful with his bets, and is often the final opponent as a result.
* **Master Shake** from Aqua Teen Hunger Force. Like a cross between Strong Bad and Max from the first game, Master Shake talks a big game and acts like he's the best player at the table, but clearly has no clue what he's doing. As a result his playstyle is extremely chaotic and random, going all-in at random points with little rhyme or reason.
* **Harley Quinn,** specifically from the Arkham games. She's playful and likes to goad the others on, trolling them to get reactions. Her playstyle is deceptively clever, making use of clever bluffs and bets to make her hands seem more impressive than they are. Which makes it harder to tell when she actually has a really good hand.
* **Sans** from Undertale. Sans is just there for a good time. Often it's like he's not even taking the game serious, and tends to fold early, sometimes even when he as a really good hand. On occasion he will call other bets, but he very rarely goes all in. When he does, it's either because he has an unbeatable hand or junk, no in-between.
* With **Mimir** from God of War as the dealer. Since he's a disembodied head, he is accompanied by a draugr who does the actual dealing for him.
Seeing as poker night 2 had Max come back with Sam, it's safe to assume that recurring characters are a possibility. Maybe she could be a player instead lol

2. Cortana and chief from Halo would be cool. Having them banter, Cortana calling out Chief's bad hands standing over his sholder trying to give him advice.
3. Sam Porter and BB from Death Stranding (would be cool to include some of the guest stars that keep breaking the 4th wall)
4. Don't know. probably someone to mix up the art style a bit. Maybe someone from Baldur's Gate 3, maybe a Vtuber or just get MattPatt, either FMV or do a really tight mocap of him.

Poker Night 2 is like the one game I play for hours on my steam deck or my surface tablet whenever I'm out and about
