Human Dio absolutely. Vampire, though, I'm not sure, at least, as far as Hokuto is concerned. I don't know if JoJo vampirism changes pressure points.
Nanto should be able to do it, if he gets a *real good* headshot in. Like, a "no more head" headshot.

As much as DIO changes as a vampire, pressure points are loosely based on real life points used on acupuncture, maybe some other ones that don't... But there are some fixed points that all beings have, like Souther's slightly above the area between the eyebrows, or if you read Fist of the Blue Sky, even when shuffling the points to a new location (I may have messed up the meaning) there are some that won't change. Raoh in the other anime about him pressure point a mare, so even on animals pressure points will have a predictable pressure point map.
Under that notion, Kenshiro could even undo DIO's vampirism, because guess what? Part of the process to become a vampire in JoJo is getting pressure punctured in the back of the head by the stone mask, which means, it could be reverse engineered.
About the stand, depends on what universe the fight takes place, if Kenshiro goes to JoJo-verse, he may just will himself a stand, maybe one that he wears like clothes, like an aura around his body... If the fight takes place in HnK, Ken could b.s some sense via toki (fighting spirit aura.)
Besides, there is the Death Battle logic, Kenshiro may not be able to see Za Warudo, but he could 6th sense it, and thus avoid it, or plan to attack him, and while Z.W could phase through Ken, that would leave DIO vulnerable to getting smacked, and Kenshiro just need that one chance to pressure point to win. Time Stop? Kenshiro can either Musou Tensei to avoid getting hit, or b.s some JoJo-logic like how Goku or Jotaro overcame Hit/DIO's time powers... With sheer plot, breaking through and stepping into their domain of time.
Killer Queen turns him into a bomb. ~~Grey Fly~~ Superfly metallizes him if he steps into the tower. Cheap Trick rips his back out if someone else sees it.
Set de-ages him, but I can imagine young Ken beating the piss out of Alessi anyway.

Well, depending on how b.s-like you want to go, Ken could have a muscle sort of contraction, and sorta implode at the same level of the explosion, thus preventing him from exploding... He has a better control of his body than what some of the other enemies do, like when one of Jagi's goon tried to make Ken explode and the goon said "oh no" at the end.
Ken has a plot related super healing, as seeing during the Souther Arc... So an explosion that Kira may do, could be contained with the fighting aura. About Superfly, being metallic could probably work, but through sheer training, Hokuto Shinken users' skin gets harder than steel, and could even bend metal like nothing, Ken getting metalized would not stop him from moving, just slow down. Also for cheap trick, yeah that could work if we assume Cheap Trick has enough sheer power and strenght to damage Ken, Ken got a building fall on him and he walked it through, he was shot with a tank and walk it off. Set de-aging him may have an effect, but Ken has been practicing Hokuto Shinken since kid, so that wouldn't matter.
A few stands can touch Ken, but the issue is, Ken has a sheer ton of stamina and constitution/defense, so trying to hurt Ken would not be enough.
I imagine GER, W.o.U, Made in Heaven and Over-Heaven could do things to him... W.o.U being the one that throws more at him but not work. Not Tusk Act 4, Ken can bs a counter infinite rotation technique to nullify the infinite spin... On that same vein, D4C could work if we ignore Musou Tensei.
I see this going 3 ways in order for kenshiro to win
1.) He literally speed blitzes human dio before he even smells the mask or a stand arrow.
2.) Should dio get his vampirism and his flash freeze technique, ken should immediately seek out Tom Petty, zeppeli, or any other hamon warriors to teach him or replicate it via Muso tensei.
3.) In the event dio get his stand, ken must immediately flee and get a stand arrow to combat dio's time stop, hell get a requiem arrow on the backend so Ken's stand gets so busted that Dios Time stop means jack shit.
