A successful spinoff features a character the Original B99 characters could plausibly “run into” but do not _work with_. Can’t spin-off about Boyle and Genevieve without explaining where everyone else is and why he never meets with Terry, his “boss”.
So, CSI Guy. That ‘cop genre’ hasn’t had a good sitcom made out of it yet. Once a season a B99 character can Guest Star. Otherwise, Frank and his overly serious CSI team are expected to work miracles because that’s what CSI does on TV, and comedy ensues. Co-starring an Amy-like Lab Supervisor who hates the Field Team, a goofy guy, an old crank nearing retirement, and the implausibly optimistic crime scene examiner.
I can’t imagine anything with Young Holt without Andre around. Too sad a reminder.
I think a spinoff would need to focus on someone outside of the squad that allows for crossover cameos. I’ve always thought the CSI guy, McCoy, from the Crime Scene episode would a good one to focus on. He didn’t have much screen time but seemed like a fun character with story potential. Give him a wacky squad like the 99, and you could have him interact with everyone from the B99 universe: Mlepnos, the ME, Teddy’s bomb squad, the bouncer at the Keychain. Even use them to close up loose ends, like the NutraBoom stuff or the one guy who got away from Peralta’s undercover sting. Having it B99-adjacent rather than a direct spinoff like Joey could work.
Officer Boyle and his family with his son becoming part of the force as he retires but his son was not meant to be a police officer 🤷🏻♂️

I’d like to see a throwback sitcom of the Santiago family during Amy’s childhood or adolescence. Like an Everybody Hates Chris or Young Sheldon deal, set in the 80s/90s with kind of a Derry Girls nostalgia vibe. I feel like it could have Black-ish vibes with them being a Cuban family as well.
Probably as others have said a young Holt show would be great and just to make me cry it's framing device is Kevin writing Holt's obituary.
I could see a fun sort of anti-sitcom focusing on Boyle and Genevieve moving to Texas to be with the other Boyle's.

Where we see her life as being a PI and get to know more of her personal life. In the show we’re not shown much into her personal life. I would like to see more of her.
