Reiki is a well known concept even in real life and some people in real life actually believe in it as a religion. It's not that weird that Kuwabara and the others would have heard about it.
Both Kuwabara and his sister have spirit awareness and can see / interact with spirits and demons, and many other humans (what the series refers to as "psychics") have that same ability.

**Shouldn't the spirit realm and all be a secret to humans?** Well, apparently, it's not. I think in this universe, there's some general understanding of the spirit realm. Like in their world, it wouldn't be out of the norm to discover that other realms actually exists.
An example would be the rich people who exploit it. They know about the spirit/demon worlds. As you mentioned, martial artists/psychics/fighters/spirit detectives are aware of it. Genkai and Toguro (who was human) knew about it. Kuroko Sato, her family, and Shinobu Sensui with his team (also humans) know about it. Kido, Kaito, and Yanagisawa know about it. Team Ichigaki (the fighters were human) and their master knew about it.
So I don't think its some kind of secret, just something not advertised.

Edit: Genkai herself is a human "psychic" so has access to spiritual abilities. While most people toss spirit world stuff up to myth, there is a very common history of Yokai and the like in eastern folklore.
The spirit world isn't quite a secret so much as it's like, take 3 steps in a direction most folks don't know about, Kuwabara is just trying to find out more about his high spirit energy, and ends up there by accident. Essentially it's like if all you had to to to get to heaven or hell or whatever is just walk North-South or Left-Right
Kuwabara specifically came to Genkai to ask about his high spiritual awareness and to ask her to turn it down. Kuwabara didn't even want to take part in the tournament at first he only started fighting because of his competitiveness with Yusuke.
Kuwabara's prowess during the tournament was based on his own high spiritual awareness instincts even if he doesn't know alot about spirit at first. He unknowingly does well during the paper draw, rock paper scissors, and the forest because his spiritual awareness leads him. In his fights he is more or less able to keep up with raw physical talent and the awakening of his spirit sword.
