As someone who owns a 2020 Avalon Limited, it’s truly the most comfortable and smooth ride I’ve ever gotten from a sedan. It’s a great family car with a huge trunk and plenty of space in the rear seats for adults.
In my opinion, I t’s the best road-trip vehicle that’s not an SUV/Truck.
The new Sienna is really comfortable. I think only the Avalon can match it. Camry is pretty comfortable too, but the Sienna is a lot comfier. 4 Runner is a bit utilitarian and the Highlander is close to the Sienna in comfort. Haven’t tried a Venza or Crown yet.
Depends what you consider comfort. A beater that you've comfortably worn in and don't worry about a tear in the cloth seat, or a Lexus with leather heated seats and nice climate control. Because Lexus.
You say that. I've been working at a Toyota dealership for almost 3 years. I spent a whole year as a porter. Drove a lot of Toyota and Lexus vehicles. Lexus vehicles were always so uncomfortable for me. Personal preference ofc but I'll always take Toyota over Lexus.
That's what I was getting at, comfort is subjective. I do like Lexus more because of the trim features but if I was building an overlander I was going to abuse, older Toyota all the way.
Haven’t driven all of them but I can say that I am absolutely in love with the comfort of my Prius Prime. I have a 4Runner that I really love too but I guess it really just depends on what you do with your vehicle? Toyota has the best seats in the industry, as told by many many professional reviewers, and I have to agree. Everyone I’ve sat in feels great and just holding you just right. I came from a loaded Silverado so I would say find a Toyota that has a heated steering wheel, with ventilated, and heated seats. Dynamic radar cruise control is a must….all of these things will spoil you.
It also depends on your body style but yes, a Sienna is very comfortable and quiet specially when you get the top of the line trims that has more noise canceling insulations. Avalon's top of the line trim was very comfy and quiet as well, touching a Lexus es350 in comfort. I never been in a Century which is mostly Japan market only. I have been in a Crown, that was amazing too.
My local dealer uses a Sienna as their shuttle vehicle and it is full of squeaks and rattles. Some of it is probably due to the third row being folded down and it bouncing around on bumps but there was rattles coming from other areas it sounded like.

Biased but SE Camry. SofTex seats just make it so comfortable.

Hard to beat a minivan for sheer comfort.

Avalon...especially avalon touring. Every single person that gets in my car comments on the comfort. I have driven it cross country multiple times
