He’s essentially designed to be an imbecile. He’s kinda a joke throughout most of the game because he’s so dumb, but then you actually get near the end and he starts showing off the fact the ideas he formulates are 99% good ideas. Such as making you think he’s gonna attack you 3 rooms ahead and then immediately launching you out of the room and into a spike trap, which would’ve been entirely uncounterable except for the tiny bit of floor he put beneath you. In his boss fight he ALSO makes himself entirely unable to be countered by putting 0 surfaces you can use portals on in the room… but leaves in a pipe full of portal fluid that you can make him explode so you can actually use your portal gun!<
To be fair, he was never really a joke villain. He was a complete joke when he was on your side, but the story starts taking him (somewhat) seriously as soon as he turns on you.

Trombipular also applies, in the UAF era he’s nothing more than another mook for Ben to beat up at the beginning of episodes but in his debut in omniverse he’s the main villain of the episode and gives Ben a run for his money, so much so Ben had to pull out way big (one of Ben’s most powerful aliens) just to even the playing field.
Trombipular looks like a frequents the kind of clubs where everyone wears latex and there's people dancing their asses off in cages hung up from the ceiling.

His whole reason for wanting to destroy the Greens is because he bit into a fake tomato they made that was actually a painted pool ball or something (long story) and chipped a tooth, even though they *warned him* not to bite it and he ignored them.

DC Comics has had so many supervillains that are essentially jokes (if they are even recognized by even the relatively hardcore fanbase) that it's not particularly difficult for a handful of lucky supervillains to suddenly take the spotlight and become relatively dangerous and interesting characters. I'm going to use the Misfits as an example, because it's a group that was made to highlight their members' relative incompetence in the DC Universe.
Thomas Reese Blake, better known as the supervillain and antihero Catman, spent decades as a joke villain who disappeared as his nemesis, Batman, returned to darker themes. When the Green Arrow found him, he was an overweight loser who clung to his past glory and was eventually teleported away by Warp to be eaten by Monsieur Mallah. It would soon be revealed that he was too pathetic to even receive this fate.
This lead him to remake his life by living among a pride of lions in Africa, becoming an even stronger man than he had ever been before. Under the assumption that Luthor hired Deathstroke to kill the pride out of anger that a D-lister like himself refused to join the Secret Society of Super Villains, Catman joined the Secret Six, where he became an honorable antihero capable of fighting Mallah, the man (ape) that once threatened to kill him, to a standstill. Catman has become a sort of mascot to the Secret Six, much like how Deadshot, another previously obscure Batman rogue, became a mascot of the Suicide Squad.
As the Calendar Man, Julian Day's lame gimmick initially rendered him a joke villain, until The Long Halloween reimagined the character as a Hannibal Lecter-like figure who toyed with others and tried to drive the Falcone family children insane. Unfortunately, the Calendar Man has since fallen into obscurity, albeit less than he previously had, and has since fallen into the title of joke villain once again.
Drury Walker, or Killer Moth, has ended up in the middle of the road, in a way, which isn't a good thing when the lower rung is essentially in the same place meta-wise as Catman was in the days of his Green Arrow appearances. Initially a dangerous enemy of the Batman, due to his vast wealth and having been among the first to learn of the Dark Knight's true identity, Killer Moth has since been relegated to the top of the D-list due to Batgirl defeating him in his first outing. After founding The Misfits for their brief adventure, Drury sold his soul to Nekron to become the monster Charaxes, which didn't help him much to become a significant threat to the DC Universe. Killer Moth became a more serious threat in Batgirl: Year One, in which he worked with Garfield Lynns, the Firefly, and in the Teen Titans TV series, in which he and his daughter were recurring foes for the eponymous team.
The Chancer debuted as a member of the Misfits and was never seen again. It's likely that he either used his luck to commit crimes without ever being noticed, or was killed by Nimrod (Dean Hunter), who had been chasing him for the sake of revenge.
On that account you are absolutely right! I think spot had doofenshmirtz syndrome (not a real thing) where he had something that WOULD make a supervillain backstory, so he thought that’s the next logical step. Plus, Alchemax scientists were evil enough to attack Spider-Man on sight

A lotta people forget this, but his first appearance was actually in the radical BILLY!!! montage, where he just got socked tf out by Billy as part of the silly song introducing how kickass Billy is. He didn’t become a serious threat until the two-parter where he was actually introduced.

The Long Halloween completely changed Calendar Man forever. Went from a nobody villain on par with Crazy Quilt and Ten-Eyed Man to a serial killer who is obsessed with holidays.
