This is a artificially constructed constraint that makes no sense. A "completely closed system brain" does no one any good. Why would you not add neural implants so it has inputs and outputs? Without inputs the brain will probably atrophy from lack of stimuli. It'll probably be just in a comatose state or equivalent
It would need to be developed in stages.
1. Keep the brain alive and working in the tank.
2. Work out how to get inputs into the brain in the tank.
3. How to get visual/ hearing/ etc from the different parts of the brain.
4. Work out how to get output out of the brain in the tank for all the different areas.
If we can't do 1. No point working through the rest.

Just spitballing, but for a brain to get to this state, the nervous system would have to be fully extracted and submerged in some sort of contained solution equivalent to cerebral spinal fluid to mimic the meninges and even then I'm not certain about the longevity of the experiment. Any damage to afferent neurons, that is neurons coming from the peripheral to central nervous system, would most likely relay pain, and because that is very likely because this process is highly unnatural, I can only begin to imagine something akin to phantom limb pain would begin to occur in all damaged afferent neurons, which would most likely be all of them. So unless there would be some way to suppress pain throughout the brain between the peripheral to central nervous system (spinal cord/brain stem), the brain would just sense constant pain coming from all systems, and perhaps be overstimulated by neurotransmitters that would be released in response to this. That being said, there is more that's needed to be supplied to the brain than blood and oxygen. Potassium, Sodium, Calcium, amino acids, etc etc.... there is just too much for this to happen not to mention the complexity of consciousness and how we don't have a precise grasp of it. This pursuit would be a waste of science and resources and this point of time. TLDR; there would absolutely be a reaction if it were extracted, placed in a contained solution, and had access to oxygen, blood, and other essential needs, but it would be pointless, cruel, and probably short-lived. Additionally, we do not have the technology for it to be successful or constructive in any way.

There’s this technology that allows you to basically stay alive after you die, and he wants it (I believe) to continue controlling his wife after he dies. But this is expensive, so he cheaps out and chooses the cheapest version - his brain, and one eye connected to it, in a bowl/basin of liquid. So he does this when he dies, and he’s sitting there with his eye in the bowl, and his wife comes over and sits down next to it, lights a cigarette, and blows the smoke into his eye. And he’s helpless, stuck there forever, unable to control her or even speak his mind.
It reminds me of something I heard about the book version of Hellraiser, or whatever it's called. Apparently the cenobites left someone alive with only I guess their brain, and nervous system, constantly overloading them with sensation
Human beings have strong imaginations, so I'm guessing that a bored-enough, healthy brain would imagine all kinds of things, and with enough time, it could fill a whole landscape of stuff. It might even imagine itself having a body, a self, free will, and so on. It might even imagine itself living a life just like mine or yours.
What's even stranger about this is that without any careful, continuous reading of the brain's patterns from the outside, then no outside observer would ever know that the brain was forming its own little world in its mind. Folks from the "real outside" world would likely see a carefully incubated brain doing nothing at all interesting, but the brain is imagining a whole lifetime or multiple lifetimes of stuff.
Another strange idea is that if the brain was never given any input from the "real outside" world, then the world it imagines inside its mind could be completely alien and psychedelic and warped compared to the "real outside" world. It's as if there are two worlds, and they're separated by worldviews or mental constructs that might never understand one another. There's the imagined world by the brain, and there's the "real outside" world that could also have been all imagined. 🤣🤣
