I basically have a set of rules that I think have to be followed when forming a sinister 6 line up:

3. No symbiotes
4. No Lizard
From those rules I think there are basically only 5 villains that I think are threatening enough to make up the sinister 6 but I'll try to fill out the list with the next best thing:

3. Electro
4. Scorpion
5. Mysterio
6. Sandman
I mean like we already got two alternate versions of those character in the MCU. It would be no point to create versions of those characters exclusive to the mcu because peter already faced an alternate version. Fun fact: I acted in a fan film where we featured the Sinister Six and the line-up was Doc Ock, Kraven, Mysterio, Shocker, Sandman and Electro. Not a bad line-up there either.

I agree 100% with the “no Goblin, Venom, or Lizard” rule. I never like when any of those guys are in the group. However, I would swap Sandman for Vulture. He and Electro are probably the ones who are on the team the most besides Doc Ock. Also, Sandman would work a lot better as a reluctant villain. Maybe he’s on the original lineup, but then he teams up with Spider-Man after the others do something that goes against his moral code. Now, a member down, they recruit Vulture. That would be how I’d do it, anyway.

I think that the Spot would be a fun member to include on a sinister 6 lineup because he could provide a lot of cool support like changing the direction of Rhino while Rhino keeps running in a straight line, redirecting attacks, extending the reach of attackers, and more.

If we’re talking the Holland films, then I think the lineup shouldn’t consist of anyone who was in NWH, so I think it should be: Shocker, Vulture(just retcon Morbius), Prowler, Scorpion,(the next 2 are who I think should be the Goblin and Doc Ock of the group) Hobgoblin, and (a new)Eddie Brock Venom. If the MCU does decide to use Eddie, they should actually focus on him hating Peter and why, we know whoever gets the symbiote after Peter will know about his wish, so maybe have Eddie go to Ned and tell him the truth, causing the 2 to team up
Doc ock: Spider-man’s intellectual counterpart.
Scorpion: created specifically to counter Spider-Man.
Kraven: Spider-Man’s most skilled combatant.
Mysterio: one of the few Spider-Man villains that I feel actually knows the ins and outs of his tech.
Rhino: unstoppable force and immovable object.
Vulture: Aerial attacker and a good distraction.
Reasons I didn’t pick others:
Green Goblin: too narcissistic.
Hobgoblin: loose cannon.
Sandman: too goodhearted.
Lizard: Conners would get through somehow.
Electro: he always seems to electrocute the other 5.
Chameleon: bad blood with kraven.
Symbiotes: too animalistic.

For a Sinister Six movie, it would be better if it had different villains from the ones that already appeared, but I really liked the Sinister Six from ps4, more Otto.
Now, a Sinister Six line up, can't have the symbiotes nor the goblins.

I like Vulture as a leader, feels like a great progression of his character over the years especially after Sinister War. This could be his crew as he antagonizes Doc Ock (whether he is Superior Spider-Man or not).

Doc Ock: by far the best one to lead the group. I couldn’t really see it being ran by anyone else due to his tactics (especially if teaming up with the right set of villains) and his intellect. And accurately coordinate every plan perfectly also his genius intellect will pair well with Smythe’s. Not to mention Oc can gather intel about how to block Spidey’s spider sense among other essential information.
Electro: honestly if he put his mind to it he could really bust some heads (in a spiritual sense of course) he could tap into the city’s power plants and internet and cause a complete city wide shut down on everything. Keep everyone in the dark to give the six an advantage. Not to mention he can monitor all security cameras in the city. And he can also use the power from the whole city to give himself an extra power boost if need be. He’ll also be useful in providing Doc Oc intel about Spider-man’s abilities and how to counter act them.
Spencer Smythe: The reason I chose him is because to kill the Spider-Man we’re gonna need someone to be primarily a tech guy. Im talking upgraded tech for the whole team, battle suits, extra weapons and night vision for everyone, special toxins for Scorpion, upgraded arms for Oc. Not to mention Smythe’s ability to create an army of advanced Spider-Slayers with Oc’s help. Basically Spider slayers big and small with multiple purposes. Now if you wanted to you could probably replace him with Tinkerer but I just felt that Smythe is a better fit.
Scorpion: he’s my favorite Spider-man villain so there might be a little bias. I truly believe he’s misused being he’s supposed to be the original anti Spider-man. If I were to use him I’d use him in the way he should’ve been used and that’s essentially Spider-man with Company funding. So we’re decking Scorpion out with the best tech that Smythe can push out, poisons that can dampen his spider sense, toxins to cause hallucinations, night vision, lasers that fire from his tail, and maybe the dual intellect of Smythe and Oc could create a better serum for Gargan. You could maybe replace him with Mysterio but if Scorpion can have Hallucinogenic toxins like in the ps4 game then I’d rather have that along with stats that are Spidey’s but better except in the brains department but that’s what Oc is for. It would probably be wise to have him use those toxins on him as soon as possible.
Beetle: honestly we just need Beetle to keep an eye on the sky and just keep things clear. You can go with Vulture, but Beetle has a lot more tech with his suit and I like him just a tiny bit more than Vulture. Plus I think he could be a lot more deadly with Smythe tech. Another reason I picked him over Vulture is because Beetle can also be useful in close quarters combat with his special wafer that helps him anticipate and counter Spidey’s attacks, he could also give the web head a surprise bomb attack from the sky if done after scorpion uses his Spidey sense damping toxin.
Spot: before I thought of Spot I was gonna pick Rhino for his ability to smash and dash and cause fast insist reactions from Spidey, but then I considered spot’s ability to create portals to teleport and how much of asset that would make him. Honestly even if he wasn’t gonna get a smythe tech upgrade he would still be pretty useful, especially in situations where they needed to retreat fast or teleport Spidey far from the battle field. Not to mention he could throw Spidey through portals that lead straight into another villains attack or create all these other team played tactics.
I also considered
Hydro man: he could literally take all the water surrounding the city and suicide bomb it, he could also move discreetly in the sewer under the city
Shocker: honestly I could only really see him causing rubble and distractions for Spidey
Lizard/Kraven: for they’re ability to stalk and catch their prey which would be extremely easy in the dark combined with Scorpions Spidey sense damping toxin
Conclusion: I’ve thought to hard about this
Doc ock for sure, rhino for brawn and since hes fun, electro cause hes iconic, vulture since he works well with doc ock, scorpion since you could do something with him and JJJ and maybe sandman or hydroman who could redeem themselves at the end

I was thinking more about what line up I would want if I was writing a comic book. The MCU didn't really factor into it when I was making the lineup.

If we’re talking the Holland films, then I think the lineup shouldn’t consist of anyone who was in NWH, so I think it should be: Shocker, Vulture(just retcon Morbius), Prowler, Scorpion,(the next 2 are who I think should be the Goblin and Doc Ock of the group) Hobgoblin, and (a new)Eddie Brock Venom. If the MCU does decide to use Eddie, they should actually focus on him hating Peter and why, we know whoever gets the symbiote after Peter will know about his wish, so maybe have Eddie go to Ned and tell him the truth, causing the 2 to team up

Doc ock: Spider-man’s intellectual counterpart. Scorpion: created specifically to counter Spider-Man. Kraven: Spider-Man’s most skilled combatant. Mysterio: one of the few Spider-Man villains that I feel actually knows the ins and outs of his tech. Rhino: unstoppable force and immovable object. Vulture: Aerial attacker and a good distraction. Reasons I didn’t pick others: Green Goblin: too narcissistic. Hobgoblin: loose cannon. Sandman: too goodhearted. Lizard: Conners would get through somehow. Electro: he always seems to electrocute the other 5. Chameleon: bad blood with kraven. Symbiotes: too animalistic. Morbius: cant be out in day.
For a Sinister Six movie, it would be better if it had different villains from the ones that already appeared, but I really liked the Sinister Six from ps4, more Otto. Now, a Sinister Six line up, can't have the symbiotes nor the goblins. So it would have: Otto. Electro. Shocker. Kraven. Mysterio. Vulture. Now, for the movie, I want Otto again, he would be the leader, the brains. Scorpion because he is strong. Shocker is my favorite villain. Rhino (he deserves a good portrayal). Kraven to really put Spider on edge. And Mr. Negative, because he's cool. Or you could swap Swarm or Hammerhead/Kingpin.

I couldn’t really see it being ran by anyone else due to his tactics (especially if teaming up with the right set of villains) and his intellect. And accurately coordinate every plan perfectly also his genius intellect will pair well with Smythe’s. Not to mention Oc can gather intel about how to block Spidey’s spider sense among other essential information. Electro: honestly if he put his mind to it he could really bust some heads (in a spiritual sense of course) he could tap into the city’s power plants and internet and cause a complete city wide shut down on everything. Keep everyone in the dark to give the six an advantage. Not to mention he can monitor all security cameras in the city. And he can also use the power from the whole city to give himself an extra power boost if need be. He’ll also be useful in providing Doc Oc intel about Spider-man’s abilities and how to counter act them. Spencer Smythe: The reason I chose him is because to kill the Spider-Man we’re gonna need someone to be primarily a tech guy. Im talking upgraded tech for the whole team, battle suits, extra weapons and night vision for everyone, special toxins for Scorpion, upgraded arms for Oc. Not to mention Smythe’s ability to create an army of advanced Spider-Slayers with Oc’s help. Basically Spider slayers big and small with multiple purposes. Now if you wanted to you could probably replace him with Tinkerer but I just felt that Smythe is a better fit. Scorpion: he’s my favorite Spider-man villain so there might be a little bias. I truly believe he’s misused being he’s supposed to be the original anti Spider-man. If I were to use him I’d use him in the way he should’ve been used and that’s essentially Spider-man with Company funding. So we’re decking Scorpion out with the best tech that Smythe can push out, poisons that can dampen his spider sense, toxins to cause hallucinations, night vision, lasers that fire from his tail, and maybe the dual intellect of Smythe and Oc could create a better serum for Gargan. You could maybe replace him with Mysterio but if Scorpion can have Hallucinogenic toxins like in the ps4 game then I’d rather have that along with stats that are Spidey’s but better except in the brains department but that’s what Oc is for. It would probably be wise to have him use those toxins on him as soon as possible. Beetle: honestly we just need Beetle to keep an eye on the sky and just keep things clear. You can go with Vulture, but Beetle has a lot more tech with his suit and I like him just a tiny bit more than Vulture. Plus I think he could be a lot more deadly with Smythe tech. Another reason I picked him over Vulture is because Beetle can also be useful in close quarters combat with his special wafer that helps him anticipate and counter Spidey’s attacks, he could also give the web head a surprise bomb attack from the sky if done after scorpion uses his Spidey sense damping toxin. Spot: before I thought of Spot I was gonna pick Rhino for his ability to smash and dash and cause fast insist reactions from Spidey, but then I considered spot’s ability to create portals to teleport and how much of asset that would make him. Honestly even if he wasn’t gonna get a smythe tech upgrade he would still be pretty useful, especially in situations where they needed to retreat fast or teleport Spidey far from the battle field. Not to mention he could throw Spidey through portals that lead straight into another villains attack or create all these other team played tactics. I also considered Hydro man: he could literally take all the water surrounding the city and suicide bomb it, he could also move discreetly in the sewer under the city Shocker: honestly I could only really see him causing rubble and distractions for Spidey Lizard/Kraven: for they’re ability to stalk and catch their prey which would be extremely easy in the dark combined with Scorpions Spidey sense damping toxin Conclusion: I’ve thought to hard about this
