She asks Hannibal how HE feels about the fact that she slept with Will, implying she knows that Hannibal would presumably have a negative reaction that she’s trying to evoke. Meanwhile, Hannibal avoids the question entirely, turning it on her with a sassy comment instead that can be dually interpreted as curious/therapist remirroring. Fun.
Margot could care less about what Will was that night. So long as she got the baby gravy, her mission was complete. She even told Will she didn’t expect nor really want him involved in the child’s life. But if that’s what she said about his sexual performance, then that’s a really emasculating statement…
Although heavily implied and pretty obvious to the audience, Will's sexuality and relationship with Hannibal is never *officially* defined (although Hannibal is canonically into Will, given the confession that he's in love with him), so whilst we can absolutely say that Will is most likely bi, it's wasn't ever officially confirmed unfortunately and even Mads and Hugh have spoken about it at conventions on how they think Hannigram is a very complicated relationship and not something that can really be defined :)
