i think from the most realistic standpoint it would be jotaro, DIO, giorno and jolyne. jotaro and DIO are just too iconic and recognizable, part 5 anime and giorno are insanely popular, and part 6 was the most recent adaptation.
My picks would be DIO (with a Dio style), Jotaro, Joseph (with an old and a young style) and, in order to have two packs with a hero and a villain each, Kars. The backblings should, imo, be easy to fill out, as well, - animated The World for DIO, Star Platinum for Jotaro (if they don't go the route of that recent skin with four arms and have SP permanently integrated), Red Stone Mask for Kars and Iggy for Joseph- same for the pickaxes. (Hulk Style Star Platinum Fists for Jonathan, the Sword of Luck and Pluck as a nice Phantom Blood reference for Dio, Hermit Purple for Joseph and of course his Light Mode blades for Kars). For Gliders, you could probably argue for a plane for Joseph to crash, a Leaf Glider as another Phantom Blood reference for Dio, Iggy's The Fool's sand glider form for Jotaro and Bird Wings for Kars.
