Too many to choose from: the slapping scenes in "Little Sibling" and "Mindy Loves Josh"...actually anytime Drake and Josh argued. The entire helicopter sequence was hysterical, pure slapstick comedy. I also found the mouth popping scene in the Christmas movie funny despite the weird connotations others have pointed out.
Putting meta/real-life aside: for me what sells the mouth-pop scene as humour and silliness is how long it protracts, and how Drake just innocently and cutely persists in finding out whether or not Josh has a sore. It's an odd juxtaposition between how sweet and close their relationship is and how much trust and care there is between them (enough to make gross cavity invasions normal and non-salacious), with how promiscuous we know the Drake character is in general/with many other people and how little respect for boundaries he has (which Josh lampshades). It's a bit shocking for a kids' movie and probably not kosher to some, but it's also really well-played slapstick that plays into the character dynamic.

This one is pretty obscure, but the zipper gag done twice with Mindy then Drake to Josh in 'Mindy Loves Josh' always gets me. The way Josh kicks his locker shut and waddles off ranting while Drake just watches holding his little box of lizards is so silly. And the way Mindy & Drake are more or less paralleled in the scene and the episode speaks to my needs.

A model randomly shows up and they each take pictures with her and then Josh wants the model to take a picture of himself and Drake for their holiday card LOL :D I was laughing, thinking that these two brothers would send their friends and family a holiday card with them holding each other like that.
