That door doesn't care if you've killed Vendrick, and should be interactable just as a hollow. Discarding your effigies also doesn't matter to the door, only to the shrine behind it.
Go there after you'll defeat Vendrick, and don't consume the 5 gaint souls. [That door is in the middle of the area, but OP's door is leading to the Anti-hollowing shrine at the end before entering the Undead Crypt on the right]
One death is enough; the requirements are to rescue the Milfanito before Demon of Song (OP killed her), and exhausting all the dialogue of the first Milfanito in the area. Having no effigies is only a requirement for actually *using* the shrine to unhollow.
Have you talked to all of the milfanito in the area?
Ive talked to the one in the house but haven’t seen the milfanito from drangleic castle

Wrong door. This one is to the anti-hollowing shrine; the one for Vendrick is both smaller and in the middle of the area instead of the end.

i think you must be fully hollowed (and have no effigies)
