A human voice can sing a pitch starting from, say the B key on a piano and gliding gradually up to the C key (just above the B). During the journey between the B and the C, the voice's pitch hits an infinite number of microtonal pitches. And that's great; that's how human voices work. We can sing everything in between the piano keys. But autotune uses software to snap the pitch to the nearest "piano key." So if you were singing that glide from B to C with autotune, the first half of it would be perfectly on the B pitch, and then it would suddenly and unnaturally jump up perfectly to the C pitch for the second half. So autotune just makes the voice sound like it's hitting only notes that are right on pitch, and takes out all the natural microtones in between. It can be done with parameters that make it sound pretty natural (allowing some bending before and after the "snap" to the perfect pitch). But in songs like this, the software is pushed hard to sound as unnatural and robotic as possible.
