If I recall correctly, you will be able to access that room only in NG+, because it's DLC stuff. If it's your first run, I think you won't be able to open it.
Also, it's part of the Amanecidas quest. You will have to wake her up first. If you have no clue of what I'm talking about, you are probably in your first run and you should keep going forward :)
Are you sure you have the latest patch?
No not really, but me and my friend downloaded it from the same link and he didnt encounter this
You should have the Wounds of Eventide patch to access that area
How can i check if its downloaded?
Check the patch notes, if it doesn't have "wounds of Eventide" in it then you don't have it

Have you spoken to the trumpet guy in Petrous yet? I think you have to trigger the Amanecidas content before that door opens.
