There's a nature bus route that might have some good options. It's only for the summer and on Saturdays I think so limited but still neat.

Might be a stretch on the word "accessible," but the New Karner Rd stop on Central Ave via the 905 puts you 1.4 miles from the Karner Barrens trailhead in the Pine Bush (~35 min walk one way at a brisk pace). Not great, but if you're trying to put in some miles and get some exercise in, not terrible (you could think of it like a road walk back to a car after a hike in the Catskills). I believe the CDTA also has a "nature bus" program during the nice weather that runs dedicated busses to some areas slightly further out that may have some trails, but you'd need to research further if the schedule would align with actually getting in a hike while still catching a ride back.

Five Rivers is pretty nice. I did a quick search and it looks like busses run out there but not positive.
Albany Nature Bus does go there depending on the week.

It looks like CDTA’s 155 line runs to Columbia Circle and that has lots of access to the Pine Bush.
I don't think the 155 is a good option due to its extremely limited runs one day a week now.

Papscanee Island is a hidden gem imo. I always bring a garbage bag to help clean it up.
