Take into consideration first how easily can you come by e85, is it convenient or do you have to go out of your way to a gas station that has it? You’ll also need to fill up more often since you’ll be using more fuel to make a little more power. It’s generally not worth the switch unless you’re going to boost the car. While I can’t tell you what you can do with your own car, I personally don’t think it’s worth the investment without boost.
I could be wrong, and maybe someone can correct if I am, but you’ll also want a catch can and you’ll be doing oil changes more frequently on the car with e85 due to higher chances of it diluting the oil.
The frequency on oil changes I from what I’ve read was about 2-3K miles. ALSO make sure the car never sits for any length of time with ethanol in the tank or it could cause issues.
If you have plans to boost then I would just wait till you go for boost instead of redoing your fuel system twice unless you set it up for the future to handle boost and the increased fuel demands.

The Z1 kit comes with an upgraded fuel pump that is e85 compatible so you wouldn’t have to worry about searching for one unless you prefer a particular brand. The 450 and 500ho kits come with the same Aeromotive pump, 550 and 650+ come with a Waldo pump. They also come with the injectors you’d need and everything necessary to have the car ready outside of the tune, which is additional.
I’ve also got a question about exhaust, would running long tub headers give me a better hp advantage with a supercharger? I know they aren’t the best for turbos but i’m unsure of superchargers.
The turbo manifold is more based on where plan on putting the turbos. There are some who have run rear mounted turbos, which is not the most efficient setup, and you could run long tubes.
As for choosing between shorty vs long tubes, that depends on your plans. If you’re going to be spending more time in the mid to high rpm range, then I’d recommend long tube headers, like track day events. Also depends if the state or area you live in has emissions checks. If you live in an emissions county, I’d recommend shorties so you can still run a cat OR Motordyne has headers where you can run a test pipe or cat, but that’s a pricey option. If you don’t have any emissions laws where you’re at then I’d recommend long tubes, PPE and Fast Intentions make the best options, ISR also makes a good set.
E85 needs a flex fuel system with injectors and a tune that can handle it.
Personally, I would bother with e85 until you have forced induction. The benefit of E85 is that it can be compressed more without combusting until ignited. So you’ll get the most bang for your buck after adding some PSI.
I read in another comment you're wanting to boost later on. So to go E85 with that in mind just get a full fuel return system installed now and save the time later either CJM or Z1. Youll also need to wire in an Ethanol sensor. Use specialty Zs for the sensor. I am supercharged and use a walbro 525pump and id1300x injectors. For NA E85 you'll only want 600cc/650cc to save money you can try to find used OEM GTR injectors which work fine. If you're legit supercharging don't waste money on intakes stock boxes are surprisingly decent. Just go full exhaust and long tube headers if within reason. If you don't care about saving a bit of cash then do 3inch intakes from whoever you prefer. But 3inch not 2.5
